Abstract

Determination of tocopherols and tocotrienols in tobacco seed oil by a normal-phase liquid chromatography

Tocopherol and tocotrienol composition in the oil of four tobacco seed varieties fromtheRepublic ofMacedonia (NS-72, P-23,YV125/3 andDjebel 48) was determined. Oil content expressed as a yield of seed dry matter varied from382 to 492 g kg-1.Anormal-phaseHPLCmethod was optimized for the quantitative determination of tocopherols and tocotrienols in tobacco seed oil. Chromatographic separation was achieved using isocratic elution, LiChrosorb Si 60 column, 1.0 mL min-1 flow rate of hexane:dioxane:ethanol (97:3:0.1 v/v/v) asmobile phase and a fluorescence detector for effluent monitoring. The three tocopherols (á-, â-, ä-) and four tocotrienols (á-, â-, ã-, ä-) were present in all oil samples. The highest content of oil tocopherols and tocotrienols isomers is determined in Djebel 48 variety.
